DNA sequencing:

what scientists 20 years ago could only dream of

1. Maxam-Gilbert chemical sequencing and Sanger dideoxy sequencing

• Both based on introduction of strand breaks at specific nucleotides followed by high resolution acrylamide gel electrophoresis
• Many variations on these basic methods are available today. See current methods books and protocols available from manufacturers for the best discussion.

2. Sequencing by hybridization with defined oligonucleotides and other modern developments may eventually prove faster and simpler, but currently high-throughput variations of Sanger sequencing are the method of choice.
